[15456761] DNA Binding; Transactivation Mouse Strong EMSA analyses using the prototypic Bright binding site (from the murine S107 V1 promoter region) showed that recombinant murine Bright has DNA binding activity. Real time PCR assays performed using mRNA isolated from CHO cells transduced with murine recombinant Bright, and a reporter gene containing part of the murine V1 S107 family heavy chain gene and promoter sequence with Bright binding sites showed that Bright has transactivaton activity. Arid3a is a TF. The recombinant protein binds DNA that contains Arid3a elements in EMSA studies. Recombinant Arid3a also transactivates a V1 S107 immunoglobulin family heavy chain reporter construct in cotransfection experiments, as evidenced by real time PCR analysis.
