[15735663] DNA Binding Human; Other Strong ATF7-induced transactivation is mediated by hsTAF12 by Gal4-ATF7 fusion proteins in Cos-1 cells with a vector expressing each of the hsTAFs being tested.

ATF7 and hsTAF12 interact in vivo by immunoprecipitation in COS-1 cells (with antibodies against ATF7 and Western Blotting using anti-TAF12 antibodies). Chromatin immunoprecipitation (ChIP) on a human lymphoid cell line (Raji cells) using an E-selectin gene demonstrates that ATF7 or TAF12 efficiently immunoprecipitates with the E-selectin promoter region suggesting simultaneous binding of both proteins to the target promoter, in vivo.

[16164412] Transactivation Human Medium Cotransfection into HepG2 cells with an expression vector encoding ATF5 and reporter gene containing the nutrient-sensing response unit of the asparagine synthetase demonstrates that ATF5 transactivates the reporter gene.

An arginine-lysine rich basic region suggests that ATF5 has sequence-specific DNA binding but this was not demonstrated by experiments. Cotransfection of a Gal4-ATF5 fusion in HepG2 cells demonstrates that ATF5 contains an active transcriptional activation domains.

Atf5 is a transcription factor. It interacts with TAF12 (by immunoprecipitation) and binds an E-selectin promoter by ChIP assay. It activates a Gal4 reporter gene fused to a GAL4 DNA binding domain demonstrating that it contains an active transcriptional activation domain and this activation potential is enhanced when TAF12 is present. It also activates a reporter gene containing the nutrient-sensing response unit of the asparagine synthetase gene.
