[9271374] Co-activation; DNA Binding Human Strong Yeast interaction screen and GST-pulldown assay demonstrate the physical association between ATF6 and SRF. ATF6 expression vector transactivates a luciferase reporter gene containing SRE c-fos promoter transfected in Hela cells, but not the reporter gene without an SRE or with a mutated SRE. ATF6 also superinduces the SRE reporter gene in serumstarved cells, suggesting that SRF and ATF6 can interact before activation of expression. Expression of the antisense ATF6 construct reduces serum-induced expression from SRE c-fos reporter by over 60%. Alice Chou
[14765107] Co-activation; DNA Binding Human Strong Co-transfection of HepG2 cells with pCMV5-HA-SREBP2(N), together with pCI-Flag-ATF6(N) or an empty pCMV5 vector, and the 4 SRE-Luc or LDLR-Luc reporters demonstrate that the HA-SREBP2(N) induced luciferase activity of the two reporters is attenuated by the expressed Flag-ATF6. Overexpression of ATF6(N) has similar inhibitory effects on SREBP2-targeted genes. The blockade of ATF6 cleavage by BiP/grp78 reverses this inhibitory effect. GST pull-down and immunoprecipitation assays reveal that ATF6(N) is bound to SREBP2(N). Deletion analysis of the various functional domains of ATF6 indicates that the interaction was through its leucine-zipper domain. Chromatin immunoprecipitation assays reveal that ATF6(N) forms a complex with the SRE-bound SREBP2(N). Alice Chou

Reviewer: Alice Chou

ATF6 and SRF can interact both in vitro and in vivo. ATF6 transactivates a SRE c-fos promoter-reporter gene in Hela cells. ATF can interact with SREBP2 both in vitro and in vivo. Increase in ATF6(N) suppresses SREBP2(N) transcriptional activity. Chromatin immunoprecipitation assays reveal that ATF6(N) forms a complex with the SRE-bound SREBP2(N).
