[11585931] TF PPI Human Strong TBP-binding regions in human TAFII170 (BTAF1) are identified. The amino terminus of hTAFII170 is found to interact with hTBP through deletion mutation analysis in a yeast two-hybrid system. Results are confirmed in a similar studies with Flag-tagged hTAFII170 and GST-hTBP, as well as coimmunoprecipitation of the endogenous proteins in COS-7 cells. These and further experiments support a mechanism in which TAFII170 induces high-mobility DNA binding by TBP through reversible interactions with its concave DNA binding surface. Amy Ticoll

Together Btaf1 and the TATA-binding protein (TBP) are the major components of TFIID (see PMID:14557059 for Review). The amino terminus of hTAFII170 is found to interact with hTBP through deletion mutation analysis in a yeast two-hybrid system. Results are confirmed with GST pull-down and coimmunoprecipitation assays.
