[16678093] Co-activation; TF PPI Human; Rat; Mouse Medium COS cells transfected with a CAMTA2 expression plasmid and ANF promoter mutant luciferase reporters demonstrate that the Nkx2-5 binding site in the ANF promoter is required for maximal transcriptional activation by CAMTA2.

Chromatin immunoprecipitation (ChIP) assays with primary neonatal rat cardiomyocytes demonstrate that CAMTA2 associated with the NKE in the ANF promoter in native chromatin. CAMTA2 fused to the GAL4 DNA binding domain activated a GAL4-dependent luciferase reporter in transfected COS cells acted as a transcription activation domain (TAD). Coimmunoprecipitation assays demonstrate that CAMTA2 interacts with HDAC5 and deletion mutants demonstrates that the interacting domains are the ankyrin-repeat domain of CAMTA2 and the N-terminal regulatory region of HDAC5.

CAMTA2 appears to act as a coactivator for Nkx2-5 on the ANF promoter by ANF promoter mutational reporter gene analysis and ChIP assay. CAMTA2 can associate with HDAC5 by coimmunoprecipitation assays, resulting in repression of CAMTA2 transcriptional activity.
