In ChIP experiments using early erythroid progenitors and late normoblasts from mouse liver, ETO-2 interacted with DNAse hypersensitive sites upstream of the mouse alpha-globin gene (known SCL targets), either alone (both cell types) or in complex with SCL (just early erythroid progenitors).

Further cotransfection of an expression construct encoding Cbfa2t3h (referred to as ETO-2 in this PMID) led to a dose-dependent decrease in reporter activity.

An interaction between ETO-2 and specific TAL-1 complex components E2A and HEB was observed in GST pulldowns in-vitro. An interaction between ETO-2 and each of the TAL-1 complex components except for GATA-1 was also demonstrated by co-immunoprecipitation from nuclear extracts of MEL cells.

ETO-2 and all other TAL-1 binding partners from TF-1 cell nuclear extracts are found to bind to the human GPA promoter immobilized on beads.

Cbfa2t3h negatively regulates the transcription of target genes in association with multiprotein complexes. Cotransfection of Cbfa2t3h with members of the SCL-containing complex into rat NIH-3T3 cells leads to a decrease in luciferase reporter expression relative to the SCL-containing complex alone, which in the absence of Cbfa2t3h brings about increased reporter gene expression. Similar results are also obtained in cotransfection experiments with all members of the TAL-1 containing protein complex. As well, ChIP experiments indicate that Cbfa2t3h interacts with DNAse hypersensitive sites upstream of the alpha-globin gene in mouse erythroid progenitor cells, both alone, and in complex with SCL.
