[12037670] Co-activation Mouse Medium Ccnt2 associated with cdk9 enhances the transcriptional activity of MyoD (gene reporter assay with the MyoD-dependent promoter 4RE-luc in NIH3T3 cells).

The kinase activity of cdk9 is necessary during muscle differentiation (C2C12 stable cell lines with different cdk9 mutants and gene reporter assay with the MCK-luc reporter in those cell lines). The N-terminus of cdk9 directly interacts in vitro with the bHLH domain of MyoD (GST-MyoD pull down with an in vitro translated cdk9). Cdk9, MyoD and cycT2 form a multimeric complex in vivo (co-IP in C2C12 cells in growth or differentiation medium). Cdk9 phosphorylates MyoD in vitro and its kinase activity peaks in the early phase of the differentiation program (in vitro kinase assay).

CycT2 only affects rates of elongation of transcription (Its effect on transcription is inhibited by PIE-1, a global inhibitor of transcription). CycT2 binds the CTD of RNAPolII in vivo (GST capture assays using Raji B cells that express stably HA epitope-tagged large subunit of RNAPII containing different lengths of the CTD).

ccnt2 is one possible regulatory subunit of the complexe P-TEFb. P-TEFb contains Cdk9 and a C-type cyclin (CycT1, CycT2a, CycT2b or CycK). Cdk9 in P-TEFb phosphorylates the C-terminal domain (CTD) of RNAPII. Ccnt2 has been shown to bind directly to RNAPII and this interaction is key for its transcriptional effects. It is also part of a complexe with MyoD, involved in MyoD transcriptional activation. It was shown that Ccnt2 can activate transcription when tethered artificially to DNA. It is possible though that the involvment of ccnt2 is only through the kinase activity of cdk9 which would give it more a role in signalling than in transcription itself.
