A luciferase-reporter assay (a promoter containing four C/EBP sites upstream of a minimal alkaline phosphatase TATA element) and deletion of the C/EBP site in the immunoglobulin VHl promoter demonstrate that endogenous Ig/EBP is not a transcriptional activator. Transfection of a luciferase-reporter construct with or without four C/EBP sites cloned upstream of a 200 bp fragment of the herpes simplex virus TK promoter in 18-81 cells (TK200Luc has easily detectable basal activity in 18-81 cells) demonstrate the inability of endogenous Ig/EBP to activate transcription. Luciferase-promoter assays involving cotransfection of Ig/EBP expression plasmids in 18-81 cells reduce the activity of the reporters suggesting that Ig/EBP inhibits activation by two C/EBP family activators, NF-IL6 and C/EBPax.

[15833715] Other Human; Mouse Medium Gel-shift assays with a radiolabelled wild-type human β-globin CCAAT-box probe and nuclear extracts from DMSO-induced MEL cells (a murine erythroleukemic cell line) with C/EBP-gamma antibodies confirm the ability of C/EBP-gamma to bind the CCAAT-box probe.

Co-transfection of plasmids encoding EKLF (EKLF is required for high-level β-globin expression during definitive erythropoiesis), C/EBPδ or C/EBPγ with a 424-bp fragment of the proximal human β-globin promoter driving expression of a luciferase reporter demonstrate C/EBPγ partially inhibits the basal activity of the reporter.

C/EBP-gamma is possibly a transcriptional inhibitor. Gel shift assays demonstrate binding to the CCAAT-box probes and luciferase-reporter assays demonstrate that C/EBP-gamma inhibits the transcriptional activities of two proteins, C/EBPax and NF-IL6.
