Supershift experiments using co-incubation of antibodies against CREB family members and nuclear extracts from 3T3-L1 preadipocytes (after exposure to the adipocyte differentiation inducers (MDI)) identify binding of CREB and ATF1 proteins to C/EBP-beta binding elements. Co-transfection of 3T3-L1 preadipocytes with a C/EBP{beta} promoter-luciferase construct and expression vectors for CREB, CREM{tau}, or ATF1 demonstrate that CREB transactivates the C/EBP-beta promoter.

[15054091] Other; DNA Binding Mouse Strong Gel-shift assays and competition experiments using nuclear proteins from N2A cells and an oligoprobe encompassing an AP-1 binding site from the murine GLUT gene promoter with the presence of anti-CREB antibody demonstrate CREB binding.

Co-transfection experiments employing a CREB expression vector and the -203-bp GLUT 3-Luciferase reporter construct were unsuccessful because of high levels of endogenous CREB present in most cell lines investigated. However, SiRNA against the CREB sequence (confirmed by Western analysis) and the -203-bp GLUT 3 promoter-luciferase reporter DNA depletes the activation of the reporter gene.

Creb is a transcription factor. It transactivates a C/EBP-beta reporter gene and siRNA against the CREB sequence depletes the activation of the GLUT 3 reporter gene.
