Multiple Crem isoforms containing two distinct DNA-binding domains are expressed by alternative splicing. All bind to the cAMP-response element (CRE: consensus TGACGTCA) in gel shift assay. Full-length Crem antagonizes Creb-induction of the somatostatin CRE when transiently cotransfected into F9 cells. Fusion proteins containining Crem deletion mutants linked to the GAL4 DNA-binding domain transactivate the GAL4-CAT reporter in transiently transfected JEG-3 cells. In similar experiments, different alternatively spliced Crem isoforms were transiently transfected into JEG-3, HeLa and F9 cells to characterize a minimal Crem repressor domain containing the bZip motif.
