Colocalization studies show that SMIF can translocate to the nucleus following treatment with BMP4 and TGF-beta and that this translocation is dependent on SMAD4. A fusion protein containing SMIF linked to the Lex DNA-binding domain activated transcription in yeast. A Gal4-SMIF fusion protein demonstrated TGF-beta, SMAD4 and CBP/p300-dependent transactivation.

No evidence found for DNA binding, but this gene is involved in transcriptional regulation through its interactions with SMAD4.
