[15464984] Co-activation; TF PPI Mouse; Unknown Strong DDX5 (p68) enhanced CBP transcriptional activity, leading to increased reporter expression, in cotransfection assays in which Mink lung epithelial (Mv1Lu) cells were transfected with a (Gal4)2-tk80-Luc reporter construct and expression constructs encoding p68, Smad3, and Gal4-CBP. Cotransfection of Smad 3 and p68, and Gal4-CBP led to more of an increase in reporter activity than just Smad3 and Gal4-CBP.

DDX5 (p68) was also shown to interact directly in-vivo with Smad3 by co-immunoprecipitation from Mv1Lu cells.

[15657085] Non-specific DNA Mouse Strong DDX5 (p68) bound to mouse centromeric minor satellite repeats (MiSats) as demonstrated in a gel mobility shift assay (GMSA) in which labeled MiSats were incubated with mouse liver nuclear extracts or nuclear matrix extracts and then subjected to gel electrophoresis. Column chromatography followed by mass spectrometry was performed using the mouse nuclear matrix extract, which bound to the probe most strongly in order to identify DDX5 as one of the proteins binding to the labeled probe. DDX5 is likely a DNA helicase which interacts with Smad3 and binds to centromeric minor satellite repeats in order to promote transcription. DDX5 increased reporter expression in complex with Smad3 in cotransfection assays while DDX5 was identified as a protein bound to mouse centromeric repeat regions through gel mobility shift assays and column chromatography followed by mass spectrometry.
