DTX1 negatively regulates the transcription of downstream targets likely in association with other cofactors. Cotransfection of E47 and DTX1 into NIH-3T3 cells leads to decreased reporter gene expression relative to E47 alone, for which there is an increase in reporter gene expression relative to reporter plasmid alone. Cotransfection of MASH1 and either of DTX1 or Notch-1 also leads to a decrease in gene expression relative to MASH1 alone, which is associated with an increase in reporter activity. As well, cotransfection of DTX1 into NIH-3T3 cells with Notch-1A and an expression construct including the mouse Hes-1 promoter leads to a decrease in reporter gene expression, while Notch-1A alone triggers an increase in reporter activity relative to reporter alone.
