[10786636] TF PPI Rat Strong NRBF-2 is identified by yeast 2-hybrid screening as interacting partner of PPAR alpha and other NRs. Transfection of yeast cells with the fusion protein of NRBF-2 with the yeast GAL4 DNA binding domain leads to reporter activation. Andrew Kwon
[15610520] TF PPI Human Strong Yeast 2 hybrid screen using the ligand-binding domain of human PPAR-alpha shows that COPR1 interacts with PPAR-alpha. COPR2 is an alternative splice variant of COPR1 and homologue of rat NRBF-2. Fusion products of COPR1 and COPR2 and their truncated versions with GAL4-DNA binding domain shows the presence of auto-activation domain. COPR1:PPAR interactions yield much higher reporter expression than COPR2:PPAR. Mutation of NR box on COPR1/COPR2 leads to decrease in reporter activity. Mutation of the AF-2 region of RXRalpha leads to decrease in reporter activity in yeast, which is also supported by mammlian two-hybrid studies. NRBF-2 is a co-transcription factor, which binds to NR via its NR box, which interacts with the AF-2 region of NR, as shown by yeast 2-hybrid assays and mutation studies.
