The Ewing's sarcoma (EWS) proto-oncogene generates EWS fusion proteins upon chromosomal translocation. The EWS protein itself is shown to bind to transcription factors and basal transcription machinery. Coimmunoprecipitation experiments demonstrate that EWS forms a complex with CBP and the homologous p300 protein, as well as TFIID and Pol II. In luciferase reporter gene assays, promoters of the human c-fos proto-oncogene, the Xenopus homeobox Xvent-2 gene, the human ErbB2 (HER2/Neu) gene and the Smad protein-dependent 3TP-lux reporter gene are stimulated by EWS.
