[9702197] Co-activation; DNA Binding; TF PPI Mouse Strong In HepG2 cells without endogenous FAST activity, gsc-Lux construct showed expression only when cotransfected with all of FAST2, SMAD2, and SMAD4. Constitutive interaction between SMAD2 and FAST2 was detected using anti-Flag Abs. In transfected COS-1 cells, EMSA with gsc element showed shift in mobility only in FAST2 expressing cells, but not in cells with SMAD2/4 only. Bacterially expressed FAST2, but not mutant FAST2, also showed shift in mobility. DNaseI footprint analysis revealed a 15 bp gsc upstream region that FAST2 binds to. Mutation of this region abolished FAST2 binding and gsc-Lux expression, confirmed by EMSA. Mutating the nearby SMAD4 binding site decreased the responsiveness of gsc promoter. Andrew Kwon
[9858566] DNA Binding; TF PPI Other Medium Xenopus FAST-1, which shares sequence homology with forkhead/HNF3beta family of TFs, binds to a 50-bp ARE element in the Mix.2 promoter as part of an ARF complex. Northern blot and RT-PCR indicates that FAST-1 expression coincides with Mix gene expression. EMSA performed with both Myc-tagged and untagged FAST-1 mRNA in frog embryo showed that it bound only to the wild type ARE motif. Antibodies against Myc tag resulted in a shift in the mobility of ARF complex, and overexpression of FAST-1 enhanced ARF formation. FAST-1 crosslinked with ARE showed the same migration pattern and similar size to ARF's major DNA-binding component crosslinked with ARE. ARF-ARE shows lower mobility, indicating additional components of ARF, and XMAD2 in conjunction with activin stimulation results in supershift of the ARF complex. Andrew Kwon

FAST2/FOXH1 appears to be a transcription factor. It binds specifically to gsc upstream region in cotransfection studies, supported by EMSA and DNaseI footprint assay. It forms a complex with SMAD2 and SMAD4, and SMAD4 is required for efficient expression of gsc gene, as shown by reporter expression studies using mutated SMAD4 binding site. SMAD2 and SMAD4 by themselves cannot bind DNA.
