[14744868] DNA Binding; Transactivation Human Strong Targets of BARX2 were identified by chromatin immunoprecipitation (ChIP) from the human MCF7 breast cancer cell line. Binding of BARX2 to 21 genomic loci examined was confirmed quantitatively by replicate ChIP assays. Electrophoretic mobility shift assays (EMSA) revealed homeodomain binding sites within several fragments that bind to BARX2 in vitro. The majority of BARX2 binding fragments tested (14/19) affected transcription in luciferase reporter gene assays. Inhibition of BARX2 expression led to reduced expression of eight genes with associated with BARX2 binding sites. Shannan HoSui

This study highlights the role of Barx2 as a repressor in the skin that can functionally interact with two WD40-domain factors distantly related to the yeast corepressor Tup1. Binding of GST-Barx2 was confirmed using EMSA. Barx2 was also detected on the region encompassing the repressor element using ChIP, as were TLE corepressors. BARX2 is a homeobox transcription factor that influences cellular differentiation in various developmental contexts. Sequence specific DNA-binding has been confirmed by ChIP and EMSA. Under different contexts, BARX2 may act as a repressor or activator of transcription.
