[12011431] DNA Binding Human Medium In a pull-down assay, ATR was shown to preferentially bind UV-damaged DNA over unmodified DNA when human Flag-tagged ATR was purified from HEK293T cells and incubated with labelled UV (6-4) photoproduct DNA or the unmodified 46bp oligomer. In vitro kinase assays carried out in the presence or absence of UV-damaged DNA showed that recombinant human ATR can phosphorylate recombinant human p53 and that the phosphorylation of p53 by purified ATR is enhanced by damaged DNA. Atr is a TF gene candidate. The endogenous human protein was found to coprecipitate with members of the nucleosome remodeling and deacetylating complex, namely CHD4, HDAC2, MTA1, and MTA2, in immunoprecipitation studies. Recombinant human ATR was shown to preferentially bind UV-damaged DNA in pull-down assays. Kinase assays showed that the recombinant protein can phosphorylate recombinant human p53, and that this phosphorylation is enhanced by damaged DNA. Current literature does not prove a transcription function for Atr, but rather indicates that Atr is involved in DNA repair. Seems to be an ssDNA damage binding protein (PMID 12011431). WWW
