[16767079] DNA Binding Human Strong Data shows that recognition of Myc-binding sites is based on epigenetic features. Euchromatic islands, defined as clusters of specific methyl- and acetyl-histone marks at the 5' end of active or poised genes, are required for Myc to bind DNA. E-boxes outside of euchromatic islands are not significantly bound, whereas virtually all E-boxes within euchromatic islands are bound as demonstrated by quantitative chromatin immunoprecipitation, methylation-sensitive restriction enzyme digestion, and bisulphite genomic sequencing. Myc is a transcription factor. It transactivates a cad-reporter in a heterodimer Cyclin T1 and an hTERT-reporter in a Myc-Max-p300 complex. Myc binds E-box motifs within euchromatic islands by quantitative chromatin immunoprecipitation, methylation-sensitive restriction enzyme digestion, and bisulphite genomic sequencing.
