[8631881] DNA Binding; Transactivation Rat Strong The primary amino acid sequence of the NZF-1 protein (Myt1l) suggested that the protein would contain two clusters of zinc fingers capable of serving as independent DNA binding domains. Competitive EMSA using bacterially expressed glutathione S-transferase fusion protins demonstrated that specific binding of NZF-1 to the beta-RARE DNA element requires a minimum of two zinc fingers. Mutations altering this element revealed that the GAAAGTT motif is critical for binding by two zinc fingers.

NZF-1 also contains regions such as acidic and serine/threonine-rich sequences commonly found in the the activation domains of many transcription factors. The potential transcriptional effects of NZF-1 were examined in CV-1 and HeLa cells, which do not contain detectable NZF-1 mRNA by RNase protection assay. NZF-1 has been shown to bind beta-RARE and Pit-1 enhancer elements in a sequence specific manner. Cotransfection of NSF-1 (under the control of the RSV promoter) and hRAR-beta promoter-luciferase reporter plasmid showed that NZF-1 moderately activated the hRAR-beta promoter ~3-fold. Cotransfection with a Pit-1 enhancer luciferase reporter in HeLa cells showed that NZF-1 activated the reporter 4-fold.

The myelin transcription factor 1 (Myt1) family represents neural zinc finger proteins of a structurally novel CCHHC class that were originally cloned by their binding to the promoter of the most abundantly expressed myelin gene, proteolipid protein. The consensus DNA-binding domain of the Myt1 family is Cys-X4-Cys-X4-His-X7-His-X5-Cys (also termed CCHHC domains). Myt1-Like (Myt1L/neural zinc finger(NZF)-1/Png-1) has been shown to bind to DNA is a sequence-specific manner by competitive EMSA. Transactivation has also been shown by luciferase reporter assays.
