[12122012] Co-activation; DNA Binding; Transactivation Human Strong Nurr1 transactivates the expression of a luciferase reporter gene containing three canonical nurr1 binding sites in PC12 cells. Cotransfection of nurr1 and RXR isotypes further demonstrates that Nurr1 interacts functionally with RXR Isotypes alpha and gamma. EMSA shows Nurr1 binds to the NBRE sites present in the 3x element and nurr1 forms DNA-bound heterodimers with RXRalpha and gamma. Alice Chou
[15485875] DNA Binding; Transactivation Rat; Mouse Strong Nurr1 protein from primary mouse osteoblasts binds to a consensus NBRE and proximal NBRE-like site in the Ocn promoter (EMSA). Nurr1 induced a consensus 3xNBRE-luciferase reporter construct in mouse osteoblasts. Ocn promoter-luciferase constructs lacking or containing a mutated proximal NBRE-like site blunts responses to Nurr1 overexpression. Nurr1 protein binds to the proximal NBRE-like site in chromatin immunoprecipitation assays. Alice Chou

Nurr1 transactivates a luciferase reporter gene containing three canonical nurr1 binding sites in PC12 cells. Cotransfection of Nurr1, RXR isotypes, and the reporter further demonstrates that Nurr1 interacts functionally with RXR Isotypes alpha and gamma. Nurr1 interacts with RXRalpha and gamma in vitro. Nurr1 directly binds to Ocn in vitro. Nurr1 transactivates a consensus 3xNBRE-containing Ocn promoter -luciferase reporter construct in mouse osteoblasts. Nurr1 protein binds to the proximal NBRE-like site in chromatin immunoprecipitation assays.
