[9418884] Co-activation; DNA Binding; TF PPI Human Strong Snapc4 (referred to as SNAP190 in this PMID) co-immunoprecipitated with SNAP45, another SNAPc complex subunit, in a co-immunoprecipitation assay in which SNAP45 in-vitro translated in reticulocyte lysate was immunoprecipitated using anti-SNAP45, and Snapc4 was detected through Western blotting using anti-Snapc4. SNAP45 also co-immunoprecipitated with Snapc4 in a reciprocal experiment.

A mobility shift, representative of Snapc4 (referred to as SNAP190 in this PMID) interacting with the proximal sequence element (PSE) of the RNA Polymerase II U6 SnRNA promoter was observed in an in-vitro EMSA assay in which in-vitro translated SNAPc complex was incubated with a labeled probe corresponding to the PSE. The presence of Snapc4 within the complex bound to the PSE was verified through a Supershift assay using anti-Snapc4.

A sharp decrease in transcription from the U1 and U6 snRNA promoters was observed in an in-vitro transcription assay as a result of the progressive depletion of Snapc4 from HeLa whole cell extracts using anti-Snapc4, and was reversed through the addition of SNAPc to immunodepleted cell extract.

Snapc4 (referred to as SNAP190 in the PMID) regulates the transcription of snRNA genes by RNA Polymerase II and III as a part of the SNAPc multiprotein complex. In-vitro translated Snapc4 co-immunoprecipitates with in-vitro translated SNAPc subunit SNAP45. However, Snapc4 does not co-immunoprecipitate with SNAPc subunits SNAP50 and SNAP43 (data discussed but not shown in the PMID). Snapc4 binds to the proximal sequence element (PSE) of the U6 SnRNA promoter transcribed by RNA Polymerase II as demonstrated by EMSA assay and verified by supershift assay. Removal of Snapc4 from HeLa whole cell extracts by immunodepletion leads to significant decreases in transcription from U1 and U6 snRNA promoters as demonstrated in an in-vitro transcription assay.
