[2493990] DNA Binding Human Strong TCF3 was cloned by Southwestern blotting from a BJAB human B-cell lymphoma cell library that was screened using a probe containing the immunoglobulin kappa chain enhancer kappa-E2 element.

Bacterially expressed TCF3 bound to the kappa-E2 element in gel shift assays, which showed that it bound DNA as a homodimer. Base-specific contacts were confirmed by methylation interference studies.

Specific E boxes that were responsible for Tcf3-dependent activation of SHP were identified using SHP promoter deletion constructs. Note that the murine homolog for this protein is Tcfe2a while the human homolog of mTcf3 is TCF7L1 (also called HMG-box transcription factor TCF-3). Human TCF3 was cloned by Southwestern blotting from a BJAB human B-cell lymphoma cell library that was screened using a probe containing the immunoglobulin kappa chain enhancer kappa-E2 element.
